Abstract摘要: 以1, 2苯并3, 4二氢咔唑9乙基氯甲酸酯( BCEOC )作为柱前衍生化试剂, 采用梯度洗脱实现了传统中藏药材小秦艽中20种氨基酸衍生物的基线分离, 利用电喷雾离子源( ESI Source) 正离子模式, 实现了小秦艽中氨基酸的定性测定, 可为秦艽类药用植物资源化学成分的深入开发利用提供依据。结果表明, 所建立的方法线性范围宽、重复性较好, 大多数氨基酸衍生物的线性回归系数大于0. 9990, 检测限为6. 5~ 178. 2 fmo l。小秦艽中氨基酸组成丰富, 至少含有18种氨基酸和7种人体必需氨基酸, 营养价值较高。同一氨基酸成分在不同采样点表现出不同含量, 体现了小秦艽该类成分的地理分布差异特征。氨基酸成分与生态环境的相关分析结果显示只有纬度与Lys之间达到了极显著的负相关关系, 其余各因子之间均未达到显著水平, 说明纬度对小秦艽氨基酸含量有较大的影响。 Abstract: A simple and sensitivem ethod for the determination o f 20 amino ac ids in an important Chinese and Tibetanm edicine Gentiana dahurica using preco lum n derivatization?reversed phase HPLC with fluorescence detection and m ass spec trom etr ic identification w as deve loped. 1, 2benzo3, 4 dihydrocarbazo le9e thylchlo roformate ( BCEOC) was used as the fluorescence derivatization reagent. 20 amino acid derivatives were separated on a Hype rsilBDSC18 colum n with a good base line resolution and detected with the fluorescence of which excitation and emission wave lengths of derivatives were set at 333 and 390 nm, respectively. Experimental results indicated that the established method had good repeat ability and the correlation coeffic ients for the amino acids derivatization were > 0. 9990. The detection limit range ( at single to noise of 3: 1) w as 6. 5 178. 2 fm o.l The identification of amino acid derivatives from hydrolyzed G. dahurica was obtained on the basis of ESI detection at positivemode. The composition of amino acids in G. dahurica was abundant and showed higher nutritional value, of which there were 18 amino acids and 7 necessary aminoacids for the health of people. The same aminoacid had different concentration level in different sampling locality and presented the character is tics of geographical differentiation. Correlation coefficient matrix between aminoacids and ecological environments showed that there was no significance among the rest factors except that latitude had a significant negative correlationre lationship with Lys at the level of 0. 01. Thus, at the present study, the ecological environm ent, especially the latitude,was one of the important factors influencing the chemical constituents ofherbalmedicines. The established method could be used and provided reference for the furthe r development and utilization of Gentiana species.
