Abstract本论文以我国传统中藏药材麻花艽(Gentiana straminea Maxim.)与达乌里秦艽(G. dahurica Fischer)为研究对象,在青藏高原大的环境背景下,在整个青海省内进行了深入广泛的取样调查,以二者药用部位根部为分析对象,利用液质联用等先进技术,定性定量地准确分析了4种环烯醚萜苷类成分(落干酸、獐牙菜苦苷、龙胆苦苷、獐牙菜苷)、30种游离脂肪酸、20种氨基酸;同时利用ICP-AES以及苯酚-硫酸法分别测定了14种无机元素、可溶性多糖的含量;同时利用数量处理方法,探讨了各化学成分特征的主要分布模式、相互关系以及高原环境对其影响,并比较分析了同属近源种之间主要化学成分特征的差异。主要研究结果如下: 1.建立了五类主要化学成分的定性、定量分析方法,准确、快捷,具可行性; 2.龙胆苦苷成分符合入药标准,氨基酸、脂肪酸、无机元素成分含量较高,而多糖类成分含量较低; 3.各主要化学成分特征存在一定关联,体现中医药多成分协同作用; 4.不同化学成分受不同环境因素的影响,证明环境是化学物质形成和变异的主要因素; 5.以五类主要化学成分的综合为变量的聚类结果,更为科学全面地反映药材品质的差异,为药材的品质鉴定和地道性归属提供依据; 6.两个物种在五类化学成分上具相似性,可能与其分布区域及环境有关。
Other AbstractGentiana straminea Maxim. & G. dahurica Fischer, belonging to the family Gentianaceae, were two famous herbaceous perennial medicine distributing in Qinghai-Tibetan Plateau. Traditionally they have been widely utilized by Tibetan folk people to relieve rheumatoid arthritis, and to remove damp-heat for the intensely bitter properties residing in the root and gentiopicroside was their marker compound of quality assessment. Currently, most studies of their chemical constituents were focused on the determination of marker compound gentiopicroside, however, other chemical constituents, such as other iridoid glycosides, amino acids and fatty acids etc, were not involved in. Therefore, in this study, throughout the Qinghai-Tibetan Plateau, a series of mass samplings were performed and determined by all kinds of modern analytical technologies. Using HPLC-MS analytical technologies, four iridoid glycosides, including loganic acid, swertiamarin, gentiopicroside and sweroside, 20 amino acids, 30 fatty acids were simultaneously determined, respectively; 14 chemical elements were also analyzed by ICP- AES; Contents of dissoluble polysaccharide were determined by the method of phenol-ritriol oil. Based on the analysis of the above described chemical constituents, the main chemical distribution pattern, relationships among all these analytes and effects of ecological environment on the distribution pattern were discussed by different statistic method, respectively. Variations between the chemical characterization of G. straminea and those of G. dahurica were also compared. The main results were listed as follows: 1.The quantitative and qualitative analytical methods of five main chemical constituents were set up, which were simple, accurate and feasible; 2.Determination of five constituents indicated that contents of gentiopicroside in G. straminea & G. dahurica were all fit for the standard using as medicine, amino acids, fatty acid and chemical elements showed high contents level, while cotents of dissoluble polysaccharide were lower 3.There was certain relationship among the determined five constituents, which showed the cooperated function among the multi-constituents. 4.Different environmental factors had different effects on different constituent, which further proved that environments were the main factor leading to the formation and variation of chemical constituents. 5.Using the integration of the determined five constituents as variables to perform the cluster analysis, it was more scientific and reasonable to control the quality assessment and guide the standardization planting. 6.There are certain comparability between the two species, which could be explained by the differentia of their different distribution.
