摘要枸杞子为茄科(Solanceae)枸杞属(Lycium L.)宁夏枸杞(Lycium bararum L.)的成熟果实,已录入《中华人民共和国药典》,具有滋补肝肾,益精明目等功效。目前,国内外针对枸杞子的研究较多,以其为主要原料的成方制剂也已在临床中得到了广泛应用。枸杞叶为枸杞(Lycium chinense Mill.)或宁夏枸杞(Lycium bararum L.)的叶,是一种传统中药。我国历代本草典籍中对其多有记载,主要具有除烦益志,补虚益精,补五劳七伤,壮心气,除热毒,散疮肿,除风明目等功效;民间亦常用枸杞叶制作药膳或药饮作保健之用。现有的资料表明:枸杞属植物的叶富含多种维生素,人体必需的氨基酸及多种矿质元素;还含有多种有用的次生代谢产物如:黄酮类化合物、萜类化合物、甾类化合物、生物碱及有机酸类等化学成分。 为了进一步明确枸杞叶中的化学成分,并对其进行有效的质量控制,本论文主要对枸杞叶的化学成分分离鉴定及其质量控制方法等做了研究,得到以下结论: 1. 分别运用有机溶剂提取法光、柱层析等方法对枸杞叶进行提取、分离,并采用紫外光谱(UV)、核磁共振谱(NMR)等方法进行化合物结构确证,结果得到两个化合物单体,其中有一个鉴定为槲皮素-3-O-α-L-鼠李糖基(1→6)-β-D-葡萄糖苷(芦丁),另一个化合物结构尚在进一步鉴定中。 2. 对枸杞叶及菠菜、海带中的矿质元素含量分析比较发现,前者Ca、Mg、P、Fe、Zn等元素的含量明显高于后两者;对枸杞叶、红鼎天精茶(枸杞叶保健茶)及普通绿茶中氨基酸含量比较显示,前两者中氨基酸含量均明显高于普通绿茶。 3. 枸杞叶药材的质量标准研究:确定了药材中芦丁的薄层鉴别条件,对其总黄酮含量测定项进行了考察,并首次建立了不同产地枸杞叶药材的高效液相色谱指纹图谱。 4. 初步建立了枸杞叶总黄酮提取物的提取纯化工艺,其中总黄酮含量达到21%以上。 5. 枸杞叶总黄酮提取物的质量标准研究:分别考察了薄层鉴别项和总黄酮含量测定项,并首次建立了枸杞叶总黄酮提取物的高效液相色谱指纹图谱。 6. 采用高效液相色谱法进行了不同海拔、不同采收期枸杞叶的总黄酮含量测定,结果显示其总黄酮含量随生长地海拔和采收期的不同均呈现一定的规律性变化,初步确定了青海枸杞叶的最佳采收期为6、7月份,最佳采集地区为柴达木地区都兰一带。
其他摘要Gouqi Zi is the fruit of Lycium barbarum L.or Lycium chinense Mill.which was recorded in Chinese Pharmacopoeia. It possesses a lot of biological activities, such as nourishing liver and kidney, immunomodulation, anti-tumor and anti-aging effect. Gouqi leaf is the leaf of L. barbarum L.or L. chinense Mill. which was recorded in many works of Chinese Meteria Medica having several efficacy such as relieving restlessness, strengthening cardiac-qi, relieving sore and tumefaction, eliminating toxic heat and improving eyesight. There are many sorts of constitiuents in leaves of Lycium L.such as amino acids, organic acids, terpenes, steroids and alkaloids. In order to confirm the constituents of L. Barbarum L. leaves and establish the quality standard of it, We studied the chemical constituents and the methods of quality control of L.barbarum L.leaves. As a result, we aquired some conclusions as follows: 1. We studied the chemical constituents of L. Barbarum L. leaves. Two compounds were aquired and one was confirmed with the methods of UV and NMR what was called to be quercetin- 3-O-α-L- rhamnopyranosyl(1→6)-β-D-glucoside (Rutin). 2. We compared the contents of amino acids and mineral elements in L. barbarum L.leaves . It was showed that the contents were higher than the other plants such as spinach, kelp and green tee. 3. We studied the quality standard of L. barbarum L. leaves including TLC- identification, HPLC content determination and HPLC fingerprint. 4. Studies on purification of total flavonoids from L. barbarum L. leaves.The content of total flavonoids is above 21%. 5. Studies on quality standard of the total flavonoids extract from L. barbarum L. leaves including TLC-identification, HPLC content determination and HPLC fingerprint. 6. The contents of total flavonoids of L. barbarum L. leaves at different harvesting times and different altitudes were determined by HPLC. It was showed that the contents were changed regularly as the altitudes and havesting time changed. The best harvesting time and best harvesting region we confirmed are June and Dulan with the altitude of 3280 m.
