唐古特铁线莲化学成分和药效学初步研究

摘要唐古特铁线莲系毛茛科铁线莲属植物,别名甘青铁线莲,藏药名又称叶芒那布、叶濛,其性辛、甘、温,具有祛寒,增生胃火,活血化淤,排脓散痈,消痞块的作用。民间常将其全草或地上部分入药,用于胃寒,消化不良,痞瘤病,黄水病及寒性肿瘤,浮肿等症的治疗。目前,从其地上部分的醇提物中分离得到了11个皂苷化合物,其中4个为新化合物。而有关其药效学的研究未见有报道。 为了寻找唐古特铁线莲更多的活性成分,本论文依以往研究为基础,利用多种柱层析分离技术,进一步对唐古特铁线莲的化学成分进行了分离、纯化和结构鉴定,从其地上部分的乙醇提取物中,分离得到4个化合物,经波谱分析(1HNMR、13CNMR、DEPT)鉴定为正壬酸,5-羟基-4-氧化-戊酸,槲皮素-3-O-葡萄糖苷,常春藤配基-3-O-α-L-鼠李糖-(1→2) -α-L-阿拉伯糖苷。其中前3个化合物为首次从该植物中分离得到。 本论文通过小鼠在体小肠运动实验和以肝癌H22型荷瘤小鼠为模型对唐古特铁线莲提取物在促进消化和抗肿瘤方面的药效学进行了初步研究。结果表明,唐古特铁线莲提取物高、中、低剂量组能推进小鼠小肠运动,其高剂量组与对照组相比,有显著性差异(P﹤0.01)。其总皂苷高、中、低三个剂量都能明显减轻肝癌H22型荷瘤小鼠瘤体重量(P﹤0.05)。 同时,本论文还运用紫外分光光度法对青海不同产地唐古特铁线莲中总皂苷的含量进行了测定。结果表明,唐古特铁线莲中总皂苷的含量与其部位有关,叶部的总皂苷含量明显高于其它部位,茎中总皂苷含量最低。同时,其总皂苷的含量还与植物生长地域有关,青海橡皮山产唐古特铁线莲中总皂苷的含量最低。本方法简便、快速、准确,可用于该药材的质量控制。
其他摘要Clematis tangutica (Maxim.) Korsh is a traditional Tibetan crude drug, known as“ye meng”, “ye mang na bu”. It tastes aigreur, sweet, and mild in nature. It is capable of disepelling cold, warming stomach, activating blood circulation to dissipate blood stasis, evacuation of pus and is used to treat stomach cold, indigestion and tumour. At present, 11 saponins were isolated from aerial part of Clematis tangutica, four of which were new triterpenoid saponins, but there is no research on its biological activities. In order to find out more effective components in it, the chemical constituents were deep studied by many kinds of chromatographic columnes on the base of fomer research, four chemical components were got from the alcohol extract of its aerial part, their structures were identified by 1HNMR, 13CNMR and DEP. They were n-Nonanoic acid, 5-hydroxy-4-oxoxpentanoic acid, quercetin-3-o-glucoside and hederagenin -3-O-α-L- rhamnopyranosyl-(1→2) - α- L –arabinopyranosoide. The first to the third of which were got from Clematis tangutica (Maxim.) Korsh for the first time. The effect of the extraction in Clematis tangutica (Maxim.) Korsh to 22 Dutch neoplasms mouse liver tumor animal models on the rate of inhibiting tumor and the impact of promoting digest by the test of ICR mouse alvine movement are discussed. The result showed that high, medium and low doses can promot mouse alvine movement, the effect was particularly prominent in the high-dose(P﹤0.01), and its high, medium and low doses can reduce the weight of neoplasms(P﹤0.05). The content of total saponins in Clematis tangutica (Maxim.) Korsh in different regions of Qinghai was determined by UV for the first time. The result indicate that the content of the total saponins in Clematis tangutica (Maxim.) Korsh relates with its parts and producting places. The content of total saponins in the leaves is more than other parts, and the content of total saponins in the Xiangpi mountain is lower than other regions. The method is simple, rapid and accurate for its quality control.
