柴达木盆地地骨皮活性成分分析方法与含量研究

摘要地骨皮是茄科植物枸杞Lycium chinense Mill.或宁夏枸杞Lycium barbarum L.的干燥根
皮。地骨皮是传统中药,现代药理学研究表明地骨皮具有多种重要的药理活性。本论文以
产于青海柴达木盆地的德令哈和都兰两个地区的地骨皮为主要研究对象,测定了地骨皮中
多种活性成分的含量,并与其它产地的地骨皮进行了比较。具体实验结果如下:
1.建立了东莨菪内酯含量测定的高效液相色谱方法,测定了青海德令哈和都兰等八个
不同产地地骨皮中东莨菪内酯的含量,其中青海两个产地东莨菪内酯的含量分别为21.00
mg/kg、10.72 mg/kg。
2.建立了五种大黄素型蒽醌化合物的高效液相色谱方法,测定了不同产地地骨皮中游
离态和结合态的五种大黄素型蒽醌化合物的含量。地骨皮中含有5 种游离的大黄素型蒽醌
化合物,结合态主要以芦荟大黄素和大黄酸为主。柴达木产地骨皮中的5 种蒽醌总量最多。
3.利用10-乙基吖啶酮-2-磺酰氯作为柱前荧光衍生试剂,建立了测定18 种氨基酸的高
效液相色谱方法,利用方法测定了不同产地地骨皮中游离氨基酸的含量。地骨皮中色氨酸
的含量最高,青海柴达木盆地两个产地地骨皮中总氨基酸的含量偏低。
4.利用2-[ 2-( 7H-二苯并[a, g]咔唑-乙氧基)-乙基氯甲酸酯(DBCEC)为柱前荧光衍生试
剂,建了测定三种牛磺酸、γ-氨基丁酸和鸟氨酸的高效液相色谱方法,利用方法测定了不
同产地地骨皮中三种非蛋白质氨基酸的含量,青海柴达木盆地两个产地地骨皮中三种氨基
酸的含量最丰富。
5.建立了测定5 种芳香有机酸的高效液相色谱方法,测定了不同产地地骨皮中游离态
和结合态的5 种芳香有机酸的含量。其中山西地骨皮中芳香有机酸的总量最大,柴达木地
骨皮中含有较多的阿魏酸。
6. 建立了测定8 种脂肪有机酸的高效液相色谱方法,测定了不同产地地骨皮中游离态
和结合态的8 种脂肪有机酸的含量。青海柴达木两个产地地骨皮中有机酸总含量几乎相同。
7. 优化了地骨皮总黄酮的提取工艺,测定了不同产地地骨皮总黄酮的提取率,并测定
了青海柴达木盆地两个产地地骨皮中总黄酮对羟基自由基和DPPH 自由基的清除能力,地
骨皮总黄酮具有较强的清除自由基的能力。
8.采用正交试验的方法优化了地骨皮多糖的提取工艺,测定了不同产地地骨皮多糖的
提取率。青海柴达木盆地两个产地地骨皮的含量分别为10.29 g/kg、13.71 g/kg。
其他摘要Cortex lycii is the dry root bark of Lycium chinense Mill. or Lycium barbarum L.. As an
important traditional Chinese medicine, the modern pharmacological studies show it has many biological activities. In this study, Cortex lyciis from Deliingha and Dulan of Qinghai province were studied. The contents of active components of cortex lycii were determined. And then a comparison was made between cortex lyciis from Deliingha and Dulan of Qinghai province and cortex lyciis from other areas. The results are as follows:
1. A high performance liquid chromatography (HPLC) method was established to
determinate the content of scopoletin in cortex lycii. Eight cortex lyciis examples, which were from different areas such as Delingha and Dulan of Qinghai province, were determined. The results showed that the contents of scopoletin in cortex lycii from Delingha and Dulan are 21.00mg/kg and 10.72 mg/kg, respectively.
2. A HPLC method was established to determinate the content of five kinds of
emodin-anthraquinone compounds in cortex lycii. The results showed that there are five kinds of free emodin-anthraquinone compounds in cortex lyciis from different areas. Aloe-emodin and rhein are the main combined emodin-anthraquinone compounds. The total content of five kinds of emodin-anthraquinone compounds are the highest of all samples.
3. Using 10-ethyl-acridone-3-sulfonyl chloride as pre-column derivatization reagent, a
HPLC method was established to determinate eighteen kinds of amino acids in cortex lycii. The content of these amino acids in cortex lyciis from different areas were determined. The results showed that the content of tryptophan is the highest in cortex lycii. The contents of all the eighteen kinds of amino acids in cortex lyciis from Delingha and Dulan of Qinghai province are lower than cortex lyciis from other areas.
4. A HPLC method Using 2-[2-(dibenzocarbazol)-ethoxy] ethyl chloroformate (DBCEC) as
pre-column derivatization reagent was established to determinate the content of taurine,
γ-aminobutyric acid and ornithine in cortex lycii. The content of these amino acids in cortex lycii from different areas were determined. The result showed that the content of these amino acids in cortex lycii from Delingha and Dulan of Qinghai province is the highest.
5. A HPLC method was established to determinate the content of five kinds of aromatic
organic acids. The content of free and combined aromatic organic acid in cortex lyciis, which were collected from different areas, were determined. The total content of aromatic organic acids in Shanxi cortex lycii is the highest of all samples.
6. A HPLC method was established to determinate the content of eight kinds of fatty organic acids. The contents of free and combined fatty organic acid in cortex lycii from different areas, were determined. There are the same level contents of total fatty organic acids in two samples from Qaidam basin.
7. The extraction technology of total flavonoids from cortex lycii was optimized. The
extraction efficiency of total flavonoids in cortex lyciis from different areas was determined. The scavenging ability of total flavonoids in cortex lycii on hydroxyl free radical and DPPH free radical was studied. The results showed that the total flavonoids in cortex lyciis have strong ability of scavenging free radicals.
8. The extraction technology of polysaccharides in cortex lyciis was optimized by using the orthogonal test. The extraction efficiency of polysaccharides in cortex lyciis from different areas was determined. The results showed that the content of polysaccharides in cortex lycii from Deliingha and Dulan of Qinghai province is 10.29 g/kg and 13.71 g/kg, respectively.
