摘要 | 五脉绿绒蒿(Meconopsis quintuplinervia Regel)是罂粟科(Papaveraceae)绿 绒蒿属(Meconopsis)多年生草本植物,系广泛使用且地位重要的传统藏药材,分 布于我国青海、西藏、陕西、甘肃和四川等地。全草或花入药,具有清热解毒, 利尿,消炎,止痛等功效,用于治疗肺炎、肝炎、头痛、水肿、皮肤病等疾病。 目前国内外关于五脉绿绒蒿化学成分的研究主要围绕于其主要活性成分的提取分 离工作,已经分离出的化学成分有:生物碱类、黄酮类、挥发油类、呋喃类等, 对于药材化学品质特征分析尚未见有研究报道。 本论文以五脉绿绒蒿作为研究对象,在青海高原大范围多点采样的基础上, 运用不同的化学分析手段,对五脉绿绒蒿中总生物碱、总黄酮、多糖、总糖、粗 蛋白、粗脂肪、氨基酸、脂肪酸、单糖以及矿物元素等化学成分含量进行了测定; 通过不同的定量化数据分析方法,得出了五脉绿绒蒿主要化学品质特征,初步建 立了适用于五脉绿绒蒿资源化学品质特征的定量化分析评价方法,并讨论了青海 高原生态环境下影响五脉绿绒蒿化学成分含量的生态因子。主要研究结果和结论 如下: 1. 各类化学成分的含量大致是以拉脊山为界,呈现出北高南低的变化趋势。 2. 对所获得的多指标定量化化学成分参数,分别采用聚类分析和模糊综合评 判两种不同方法分析五脉绿绒蒿化学品质特征。两种方法所得结果均显示出五脉 绿绒蒿化学品质总体以拉脊山为界,北高南低的特征。相比而言,模糊评判更能 直观的得出五脉绿绒蒿化学成分品质较高的产地。故选取模糊评判为五脉绿绒蒿 化学成分品质特征分析的适宜方法。 3. 通过灰色关联度分析,找出了与各类化学成分关联度较高的生态因子,即 有效磷与总生物碱、多糖含量关联度最高;纬度与总黄酮、单糖和粗蛋白含量具 有较高的关联度;海拔与总糖、有机质与氨基酸、全磷和粗脂肪、全氮与脂肪酸、 全钾与矿物元素关联度最高。而与五脉绿绒蒿化学品质关联度最高的生态因子为 纬度。 |
其他摘要 | Meconopsis quintuplinervia Regel., belongs to the Papaveraceae genus, Meconopsis family, was perennial herb distributing in Qinghai, Tibet, Shanxi, Gansu and Sichuan of P.R. China. As a widely using and important Tibetan medical material, the whole plant or flower have the analgesic, heat-clearing and detoxifying effectiveness, such as the efficacy for the treatment of pneumonia, hepatitis and other diseases. So far, studies of its chemical compounds were focused on extraction and separation of the active ingredient in vivo, like alkaloids, flavone, volatile oils, furans and others. Its chemical quality characteristic was not involved in. In this paper, we have collected M. quintuplinervia from 16 points from south to north in Qinghai plateau. Then the contents of total alkaloids, total flavone, amino acids, free fatty acids and other 6 chemical ingredients in M. quintuplinervia were determined by HPLC, ICP-OES, UV-spectrophotometry and some other instruments or methods. By comparing fuzzy comprehensive evaluation method (FCE) and cluster analysis method (CA), those data were conducted to build a suitable evaluation method to obtain the chemical quality characteristic. And also, the data were analyzed by grey relational analysis to confirm the main ecological factors that affect those chemicals and the quality. The main findings and conclusions of this research were summarized as follows. 1. The contents of all chemical composition of M. quintuplinervia were generally separated by the Laji Mountain, presenting the pattern that the contents of those tested chemicals are higher in samples from north than south. 2. CA and FCE method were applied to analyze chemical quality characteristic and to establish the analyzing method. The results of the methods both show that samples from Qunjia County and Youhulugoukou points are of a higher quality level. In other words, their chemical quality are better than the others. And the FCE have been established as a suitable method to analyze chemical quality characteristic of the Tibetan medical material, M. quintuplinervia. 3. The correlation values given by grey correlation analysis show that ecological factors are affect chemical composition in different degrees. The dominant factors are available P, latitude, altitude, organic matter, total P, total N and total K. Latitude is the main factor which affect the chemical quality the most. |
