唐古特大黄蒽醌类成分变异研究; Anthraquinones variation of Rheum tanguticum Maxim.ex Balf.

摘要大黄是我国著名的传统中药,具有泻热通肠,凉血解毒,逐瘀通经之功效。唐古特大黄是我国特有物种,中国药典(2005年版,一部)规定其为正品大黄之一,青海省是唐古特大黄地道药材的主要产区。随着大黄临床应用领域的扩展和新产品的开发,国内外市场对大黄需求量不断增长,由于掠夺式的滥采滥挖,造成大黄野生资源锐减,对产地生态环境的平衡造成破坏,并影响了地区经济的可持续性发展。因此,保护和选育优质唐古特大黄优良种质资源具有重要意义。 本研究采用HPLC法对不同产地唐古特大黄蒽醌类成分进行研究,主要研究成果如下: 1唐古特大黄主根不同部位间(根头、根身和根尾)芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚含量均无明显差异。而侧根中蒽醌化合物的含量明显高于主根。 2 青海地区唐古特大黄5种蒽醌类化合物的含量的居群内变异大于居群间变异,居群内变异是唐古特大黄表型变异的主要部分。 3 不同居群的化学成分距离矩阵和地理距离矩阵的Mental检验表明,二者的相关性较低。; “Da-Huang” (Rhubarb), a famous Traditional Chinese Medicine, has been used to befall heat, cool the blood, and clear toxic purges. Rheum tanguticum Maxim.ex Balf., as a peculiar species of our country, is recognized and recorded in the Chinese Pharmacopoeia as one of official rhubarb. Qinghai province is the main product area of genuine R. tanguticum. Natural officinal resources of R. tanguticum have decreased greatly owing to overexploitation. What is worse, the source of cultivar rhubarb germplasm is confused. The low and high quality of different R. tanguticum is inconventient to clinical application. So it is important to breed high quality germplasm resource. Anthraquinones yield in sixteen populations of R. tanguticum were determined by HPLC. The main results are as follows: The content of five anthraquinones of different parts of rhubarb (the head, the body and the part of the root) was not significant differences. There were significant differences in the contents of the five anthraquinones, the contents of five anthraquinones of the lateral root was more than the main root. Secondary metabolites are morphologic traits of medicinal plant. The variance analysis shows that variations of all the five anthraquinones are significant among and within populations, which may result from genetic and environment factors. Intrapopulation variation mainly contributes to the variation, and the phenotypic differentiation coefficient is 38.82%. The Mantel test showed there were no strong correlation between chemical composition matrix and geographical distance matrix.
