摘要 | On a reversed-phase Eclipse XDB-C-8 chromatographic column, 20 fatty acids were derivatized using 2-(2-(anthracen-10-yl)-1H-naphtho[2,3-d] imidazol-1-yl) ethyl-p-toluenesulfonate (ANITS) as precolumn derivatization reagent and separated by HPLC in conjunction with a gradient elution with fluorescence detection and MS identification. The reagent reacted with fatty acids in the presence of K2CO3 catalyst at 90 degrees C for 40 min in N, N-dimethylformamide solvent to give maximum and constant fluorescence intensity with the addition of 7-fold molar reagent excess to total molar fatty acids. The fluorescence excitation and emission wavelengths of derivatives were lambda(ex) =250 mn and lambda(em) =512 nm, respectively. The identification of fatty acid derivatives was performed by the online APCI/MS (atmospheric pressure chemical ionization source) in positive mode. All linear correlation coefficients for fatty acid derivatives are over 0. 9999, and detection limits, at signal-to-noise of 3:1, are 24. 76 - 98. 79 fmol for labeled fatty acids. The established method is sensitive and repeatable. It has been applied to for the determination of free fatty acids from rape bee pollen samples with satisfactory results. |
