摘要藜为藜科藜属植物,田园中杂草,产于全国各省区。藏医药中藜性甘﹑平,全草具有发散风热,祛风,清热的功效;用于治疗感冒等。本论文对藜叶中主要化学成分和黄酮类化合物的抗氧化活性进行了系统和深入研究,为藜资源的进一步开发利用提供科学依据。主要研究内容包括: 1.藜叶中总黄酮含量为2.43%。 2.初步建立了藜叶中黄酮类化合物的提取方法及其提取工艺条件。最佳的提取方法为醇提法;最佳的提取工艺在过40目筛和提取3次条件下为:提取温度为80℃,乙醇浓度为85%,提取时间为80分钟,料液比为1:22(g:ml),其中乙醇浓度对藜叶中黄酮类化合物的提取影响显著。 3.分别运用有机溶剂提取法、柱层析等方法对藜叶进行提取、分离,并对化合物进行结构鉴定,分离出芦丁。 4. 对藜叶中分离出的芦丁进行抗氧化活性研究,并且与Vc﹑乙酸乙酯部位浸膏和正丁醇部位浸膏清除DPPH•﹑O2-•和•OH自由基的能力进行比较。结果表明:芦丁清除DPPH自由基的半抑制浓度为0.05μg/ml,清除能力大小为:Vc﹥芦丁﹥乙酸乙酯浸膏﹥正丁醇浸膏;芦丁清除O2-自由基的半抑制浓度为80.3μg/ml,清除能力大小为:Vc﹥正丁醇浸膏﹥乙酸乙酯浸膏﹥芦丁(根据半抑制浓度);芦丁清除•OH自由基的半抑制浓度为56.8μg/ml,清除能力大小为:芦丁﹥Vc﹥正丁醇浸膏﹥乙酸乙酯浸膏。芦丁对DPPH自由基具有较强的清除能力。 5.藜叶中含有人体必需的Cu、Zn、Fe 、Mn、Co 、Ni 、Cr等微量元素。 6.藜叶中蛋白质含量为34.3%;氨基酸总量为13.7%。
其他摘要Chenopodium album L. belongs to Chenopodiacae,it is one of plant of Chenopodium plants.it is considered weed,it is widely distributing in China and abundantly in Qinghai.it is a Tibetan medicinal herb,the drug is neither cool nor warm,the flavor is sweet,it can be used in the treatment of fever and rheum.In this research, the chemical constituents and antioxidant activity of Chenopodium album L. ,have been studied widely and systematically.The results and conclusions as followed could provide scientific basis for futher research and development of its resource. 1.The content of total flavonoids in Chenopodium album L.leaves is 2.43% . 2.Constructing better extraction method of total flavonoids from Chenopodium album L.leaves and the optimal extraction technics conditions. Solvent extraction method is better,and it’s optimal extraction conditions are: ethanol concentration 85%,extraction time 80 min,material to solvent 1:22 (g:ml), extraction temperature 80℃,which is based on extraction time and the quantity ratio of material to solvent under the conditions of three extraction times and 40 screen mesh.,ethanol concentration has distinct affect. 3.We studied the chemical constituents of Chenopodium album L.leaves. Three compounds were acquired and one confirmed is Rutin. 4. Studies on antioxidant activity of Rutin from Chenopodium album L.leaves, and was compared with Vc.The results exhibited Rutin has strong DPPH radicals scaveging activity,the concentration of its IC50 is 0.05μg/ml.The sequence of scavaging DPPH• capability is:Vc>Rutin>ethyl acetate part>n-butanol part;The sequence of scavaging O2-• capability is:Vc>n-butanol part>ethyl acetate part >Rutin(according to IC50);The sequence of scavaging•OH capability is:Rutin>Vc>n-butanol part >ethyl acetate part. 5.There are body needed trace elements,such as Cu、Zn、Fe、Mn、Co、Ni、Cr et al. 6.The content of protein is 34.3%; the total aminophenol is up to 13.7%.
