中文摘要: 五脉绿绒蒿(Meconopsis quintuplinervia Regel)为多年生草本植物,隶属于罂粟科(Papaveraceae)绿绒蒿属(Meconopsis),分布于我国青海、西藏、陕西、甘肃和四川等地。五脉绿绒蒿为使用广泛且地位重要的藏药材,全草或花入药,具有清热解毒,利尿,消炎,止痛等功效,用于治疗肺炎、肝炎、头痛、水肿、皮肤病等疾病。目前,关于五脉绿绒蒿化学成分的研究主要围绕着其体内活性成分的提取分离,已经分离出的有:生物碱类、黄酮类、挥发油类、呋喃类以及其它化学成分。对于药材黄酮的含量和生态环境因子对其化学成分含量的影响未有研究。 本论文在青藏高原大的生态环境条件下,在青海省内药材主要分布区进行了深入广泛的取样调查。以五脉绿绒蒿全草作为研究对象,利用紫外分光光度法和HPLC法定量准确分析了总生物碱和木犀草素、槲皮素、山奈素及异鼠李素的含量,并计算出总黄酮的含量。采用土壤农化分析方法分析测定了采样位点土壤养分含量与PH值。在实验分析的基础上,利用统计学处理方法,探讨了生态环境对化学成分含量的影响,并初步比较分析了不同产地资源品质的高低。主要研究结果和结论总结如下: 1. 不同样品间总生物碱含量差异显著; 黄酮成分分为可以相互转化的两类:双氢黄酮类(木犀草素)和双氢黄酮醇类(槲皮素、山奈素、异鼠李素),四种黄酮类成分含量中以木犀草素和槲皮素为主,属于双氢黄酮醇类三种成分之间存在协同作用。 2. 海拔和经纬度是各种生态因子的综合体现,纬度对总生物碱和木犀草素含量的影响最明显,海拔也对木犀草素含量有显著影响。 3. 样点土壤样品的分析结果表明,当土壤中氮、磷、钾和有机质含量较高时,五脉绿绒蒿药材中总生物碱和槲皮素、山奈素、异鼠李素含量较低,木犀草素的含量较高;当土壤PH值较高时,检测样品中总生物碱和木犀草素含量较高,而其他三种双氢黄酮醇类的含量则较低。 4. 光照、温度和降水等气象因子对化学成分也有显著影响。在一定范围内,随着日照时数的增加,五脉绿绒蒿药材总黄酮成分含量增加,木犀草素含量减少。当处于低温湿润的气象条件下时,五脉绿绒蒿药材总黄酮含量较高,而处于高温干旱下木犀草素含量较高,可能是受到低温和干旱的逆境胁迫所致。 5.不同产地药材间化学成分存在差异。综合来看,达日和玛沁两县五脉绿绒蒿中的总生物碱含量较高,总黄酮及各种黄酮成分含量最高,属于综合品质较好的类群;同仁和循化两县总生物碱含量最高,但黄酮含量偏低,属于高生物碱品质的类群;祁连和民乐两县的木犀草素含量最高,其他成分偏低,属于高木犀草素品质的类群;其他地区的五脉绿绒蒿属于中在测定成分中无明显特点的类群。
英文摘要: Meconopsis quintuplinervia Regel, belong to the species from the genus Meconopsis of Papaveraceae, were Perennial herb distributing in Qinghai, Tibet, Shaanxi, Gansu and Sichuan in P R China. As a extensive and important Tibetan medicine, the whole plant or flower can be use in medicine, with heat-clearing and detoxifying, diuretic, anti-inflammatory effectiveness, analgesic, such as the efficacy for the treatment of pneumonia, hepatitis, headache, edema, skin diseases and other diseases. Currently, most studies of it chemical constituents were focused on extraction and separation of the active ingredient in vivo, alkaloids, flavonoids, volatile oils, furans and other chemical constituents have been isolated from the medicinal material. However, the content of flavonoids in herbs and eco-environmental factors on their chemical composition were not involved in. In this paper, throughout the ecological environment of the Qinghai-Tibet Plateau, an in-depth and wide range sample surveys were developed in the main distribution area of Qinghai area. With the whole plant of Meconopsis quintuplinervia as the study object, using UV spectrophotometry and HPLC accurate analysis the total alkaloids and luteolin, quercetin, kaempferol and isorhamnetin content quantity, also to calculate the total content of flavonoids. Soil nutrient content and PH value of sampling sites were determined by agricultural chemical method. Based on the experimental analysis, we explore the effects of ecological environment to chemical composition by statistical methods, and give a preliminary comparative analysis of the quality in different origin resources. The main findings and conclusions are summarized as follows: 1. Total alkaloid content between the samples was significantly different; flavonoids could be dived into two categories: flavanone (luteolin) and flavanone alcohols (quercetin, kaempferol and isorhamnetin), luteolin and quercetin occupied most in four flavonoids. The synergies were existented between three alcohols flavanone components. 2. Elevation, longitude and latitude were integrated embodiment of various ecological factors. The effects of latitude to total alkaloids and luteolin were significant, also elevation to luteolin. 3. Soil sample analysis results show that,when the soil rich in nitrogen, phosphorus, potassium and organic matter content,the contents of total alkaloid and quercetin, kaempferol and isorhamnetin with a high level in herbs, but the content of luteolin with a low level; and when soil with a high pH value, contents of total alkaloid and luteolin increase but the other three flavanone alcohols contents decrease. This may be related to internal medicine itself metabolism process. 4. Light, temperature and precipitation and other meteorological factors have a significant impact on the chemical composition too. In a certain extent, with the increase of sunshine hours, total alkaloids and total flavonoids ingredients increased, luteolin decreased. Total alkaloids and total flavonoids in herbs with a high level in low-temperature and humid weather conditions, and luteolin with a high level under high temperature and drought, but this may be also by low temperature and drought stress caused by the adversity. 5. The chemical constituents of herb between every origin are different. From the integrated quality analysis, counties of Dari and Maqin with a high level of total alkaloids, highest quality of flavonoids and the various components contained are the best integrated groups; Tongren and Xunhua both have the highest total alkaloid content, but low content of flavonoids, belong to high-quality alkaloids groups; Qilian and Minle of the highest luteolin content with other components in the low side, are high quality luteolin groups; Meconopsis quintuplinervia in other parts belonging to groups had no significant. in determination components.
