摘要 | In the present work, a new system of drug delivery to enhance the chemotherapy efficiency for cancer was established. Novel core-shell structures were presented here, which were consisted of poly (N-isopropyl acrylamide) (PNIPAM) microspheres as cores and gold as shells. Next, the chemotherapeutic agent Verbascoside (VB) was combined with PNIPAM/gold nanoshells (GNSs) structures (PMS) for inducing apoptosis in drug resistant leukemia cells (K562/A02; KA). The exploration on xenografted tumors in KA nude mice indicated that anticancer drug loaded on PNIPAM/GNSs was an efficient drug delivery vector. It induced the apoptosis of KA cells in vitro and in vivo. Apoptotic staining and DNA fragmentation further demonstrated that the apoptosis rate increased via the treatment of PMS together with VB (VB-PMS) in KA cells, compared with PMS or VB treatment alone. Moreover, VB-PMS activated the expression of apoptosis-related caspase proteins in KA cells. In summary, a novel drug delivery system has been established, and has further been proved that the combination of VB and PMS could improve therapeutic efficacy for leukemia. |
